Shimadzu: HiPLEX-IHC Imaging on an Entry-Level Benchtop MALDI-TOF Mass Spectrometer

In recent years, Matrix assisted laser desorption ionization immunohistochemistry (MALDI-IHC) has become a popular technique in imaging. MALDI-IHC uses a photocleavable peptide mass tag conjugated with an antibody to target antigens within a sample. Subsequent cleavage of the mass tag allows for this to be detected by MALDI-TOF. Using mass spectrometry imaging (MSI), spatial information of the cleaved peptide can be determined and, from this, the locations of the targets in the tissue, can be inferred.

In this webinar, we will cover some of the basics on what MALDI is and how it can be used as a tool for imaging before exploring MALDI-IHC in more detail including some examples of its implementation on our entry level 8000 series linear MALDI-TOF benchtop mass spectrometer. Our aim is to show how easy it can be to incorporate MALDI imaging into your laboratory, especially if you currently use immunohistochemistry workflows. We are ready to support you wherever you may be on your MALDI imaging journey.

We will host a Q&A session after the presentation to address any questions or comments.

Presenter: Caroline Jones (Global MALDI Applications Specialist, Kratos Analytical)

Caroline Jones graduated from the University of Leeds with a Bachelor’s degree in Genetics following which she worked on drug metabolism and pharmacokinetic studies of new pharmaceutical products using LC/MS-MS within a GLP environment. Two years later she returned to study at the University of Manchester, graduating with a Master’s degree in Immunology and Immunogenetics. Her research focused on the detection of peripheral dendritic cell concentrations using triple color flow cytometry and their potential as a predictor for post-transplant complications in patients. On graduation, she took up a position becoming a Senior Cytogenetic Technologist at St Mary’s Hospital in Manchester using techniques in cell culture and microscopy to identify and provide clinical interpretation of chromosome abnormalities in a variety of human samples. After a break from science to raise her family of 4 daughters, she returned to a position at Kratos Analytical in Manchester, UK where she is now a Global MALDI Applications Specialist. Here, she works within a team to develop MALDI-based applications to showcase the Shimadzu MALDI products with a focus on MALDI imaging. She also works alongside the R&D Department to support the development of new Shimadzu MALDI instruments and software.

Presenter: Nazim Boutaghou, Ph.D. (Life Science Product Manager, Shimadzu Scientific Instruments)

Nazim Boutaghou received a Master’s degree with a dual major in Chemistry and Biology from the University of Bordeaux 1. He joined Dr. Jean Marie Schmitter’s research group at the European Institute of Chemistry and Biology and worked on establishing H/D exchange protocols to study molecular processes taking place in the early onset of the Alzheimer’s disease. After a short break from science to study business management, Nazim joined Dr. Richard Coles’ group at the Louisiana State University in New Orleans (UNO) and advanced his work in the field of Mass Spectrometry, specializing mostly on MALDI-based workflows. He authored and co-authored several publications related to development of novel MALDI matrices, and various proteomics topics. He joined Shimadzu Scientific Instruments (SSI, American Subsidiary of Shimadzu Corporation) in 2013 as an application scientist and became product manager for the life Science products in 2019. He leads a team of several scientist that support life science products and applications for SSI.
