Automated workflow for mRNA sequencing by High Resolution LCMS

The recent successful advances with mRNA vaccines and therapeutic oligonucleotides have led to a dramatic increase in the requirement for analytical methods for their characterization. Sequence confirmation of therapeutic proteins by high resolution LCMS is an established technique to characterize and verify identity of the product. Routine sequence confirmation of mRNA using high resolution LCMS however, has not yet been established due to several challenges in the workflow. Digestion of mRNA by nuclease’s is rapid and usually creates fragments too small to be matched to a sequence. In addition, unlike peptide mapping, there has been a lack of commercially available software to annotate and match fragment MS/MS sequence data to the native full sequence.
Advances in enzymatic digestion techniques combined with new software analysis tools that incorporate in silico digestion capabilities for mRNA sequences will demonstrate the potential for sequence mapping and confirmation of mRNA using high resolution LCMS. These new advances have the potential to progress mRNA sequencing by LCMS into the same well-defined workflow currently in place for the peptide mapping of therapeutic proteins.

His current job title is the European team leader for the BioPharma/Pharma expert support group. This involves the support of Bio-Pharmaceutical applications, Involving ongoing collaborations with the Biopharmaceutical industry and academia. Method development for the characterization of biological compounds by LC/HRMS, including novel coupling to MS.

Keeley Murphy joined Thermo Fisher Scientific in 2008 and is currently working in the role of Software Product Manager, with a focus on the development of software workflows to support the LCMS analysis of oligonucleotides and mRNA. With 12 years of experience within Thermo Fisher Scientific, Keeley has contributed to numerous product launches and introductions for both hardware and software technologies designed to support biopharmaceutical development.
