Význam tématu


Tripterygium wilfordii obsahuje toxické alkaloidy Wilfordine a Wilforine, které se mohou transferovat do medu prostřednictvím včelího sběru nektaru. Nízké hladiny těchto sloučenin a složitá matice medu vyžadují vysoce citlivé a selektivní metody pro jejich spolehlivou detekci v potravinářské praxi.

Cíle a přehled studie


Cílem studie bylo vyvinout a ověřit rychlou, citlivou a reprodukovatelnou metodu pro kvantifikaci Wilfordine a Wilforine v medu. Autoři zvolili kombinaci bezvodé extrakce kapalina–kapalina (LLE) a vysokorychlostní kapalinové chromatografie s tandemovou hmotnostní spektrometrií (LC-MS/MS).

Použitá metodika


Vzorky: 1 g medu bylo naředěno vodou, extrahováno acetonitrilem s přídavkem NaCl a MgSO₄, protřepáno, odstředěno a filtrátem (0,22 μm) přímo injektováno.

Chromatografie: kolona C8 (2,1×150 mm, 5 μm), mobilní fáze A – 0,1 % kyselina mravenčí ve vodě, B – acetonitril. Gradientní program: 30 % B (1 min), 90 % B (4–5 min), návrat na 30 % B (5,1–10 min). Teplota kolonky 35 °C, objem injekce 10 μL.

Detekce MS/MS: ionizace ESI v pozitivním módu MRM, ultrarychlé přepínání polarity (5 ms), MRM přechody optimalizovány pro kvantifikaci i kvalifikaci obou sloučenin.

Hlavní výsledky a diskuse


Kalibrace byla ověřena externími standardy v rozsahu 0,01–10 ng/mL, korelační koeficienty nad 0,9996. Mez detekce (S/N=3) dosáhla přibližně 1,3 ng/L pro Wilforine a 4,3 ng/L pro Wilfordine, mez stanovitelnosti (S/N=10) 2,7 a 9,0 ng/L. Obnovení (recovery) ve spikovaných vzorcích medu (0,05 a 5,0 ng/g) se pohybovalo mezi 98,8–116,0 %, což potvrzuje přesnost a přesnost metody.

Přínosy a praktické využití metody


Navržená LLE-LC-MS/MS metoda umožňuje rychlé, citlivé a selektivní stanovení Wilfordine a Wilforine v potravinářské matrici medu. Je vhodná pro rutinní kontrolu kvality, toxikologické screeningy a výzkum bezpečnosti potravin.

Závěr


Byla vyvinuta validovaná metoda založená na LLE a LC-MS/MS, která prokázala vynikající linearitu, nízké meze detekce a kvantifikace a vysoké recovery při analýze Wilfordine a Wilforine v medu. Metoda je vhodná pro aplikace v rámci QA/QC a výzkumných laboratoří.
